Vine (UAE Team Emirates-XRG) launched a decisive attack with 20km remaining, breaking clear from an early breakaway group on the 170.3km mountain route from Olot in Catalonia to Pal in Andorra.
The 29-year-old Australian secured his third individual stage win at the Vuelta.
Norway’s Traeen, 30, finished 54 seconds behind Vine. The Bahrain Victorious rider surged from 26th to first in the general classification.
Traeen, who overcame testicular cancer in 2022 and was declared cancer-free in 2023, now leads by 31 seconds over France’s Bruno Armirail.
Italy’s Lorenzo Fortunato (XDS Astana) claimed third place in the first summit finish of this year’s race and sits third overall.
Two-time Tour de France champion Jonas Vingegaard, who started the day in the red jersey, dropped to fifth overall, 2 minutes 33 seconds behind Traeen.
Great Britain’s James Shaw (EF Education-EasyPost) finished sixth on the day and is 21st in the overall standings.
Stage seven on Friday features another mountain challenge, running from Andorra la Vella to Cerler.
